摘    要:
目的:研究膀胱平滑肌细胞与膀胱脱细胞基质(BAM)的生物相容性。方法:分离培养兔膀胱平滑肌细胞,采用α-平滑肌肌动蛋白(α-SMA)抗体进行鉴定。将以1×10 5个/mL的单细胞悬液均匀接种于制备BAM支架上,通过与单独培养的膀胱平滑肌细胞作对照,绘制两组细胞生长曲线图,对比观察两条生长曲线的差异性。 ...

Abstract:
Objective To study the biocompatibility of bladder smooth muscle cells and bladder acellular matrix(BAM).Methods Bladder smooth musrle cells were isolated and cultured,their morphological characteristics were observed under inverted microscope,and identified by α-smooth muscle actin(α-SMA)antibody.Then the 4th generation bladder smooth muscle cells were seeded on HAM scaffold with a density of 1 × 10^5 cells/mL.The separated cultured bladder smooth muscle cells were regarded as control group.The cell growth curves of the two groups were drawn and the difference between them was observed.Results The morphology of the 4th generation bladder smooth muscle cells was typical"long fusiform".Immunofluorescence showed positive expression of α-SAM.Bladder smooth muscle cells could adhere and growth on the surface of BAM,and the cell growth curves of the two groups are basically similar.Conclusions Bladder smooth muscle cells have good hiocompatihility with BAM.Bladder smooth muscle cells can be used as seed cells and BAM can be used as scaffold material to construct tissue-engineered urinary tract graft.
