摘    要:目的:探讨不同年龄成体肌肉干细胞—卫星细胞体外培养增殖、分化规律,了解卫星细胞生长能力与年龄的关系。方法:用MTT法、免疫细胞化学技术及粘附细胞仪等手段,检测不同年龄段大鼠多代肌卫星细胞的增殖、分化能力和细胞数量。结果:(1)单位重量细胞数与动物年龄呈负相关;(2)不同年龄组间细胞增殖能力从7月龄始明显递减,分化能力从11月龄始减弱。结论:卫星细胞数量与年龄呈负相关,老年大鼠卫星细胞生长能力显著减弱。

Abstract:Objective: To study the rule of proliferation and differentiation of muscle stem cells-satellite cell(SCs) from various age rats in vitro and explore the relationship between SCs and aging.Methods: The quantity and quality of SCs from various age rats were compared by MTT,immune cytochemistry and ACAS.Results: The number of SCs was of a negative correlation with the age.The activity and growing ability of SCs was very stable from 1 month old to 6 months old,but they became worse after 7 months old and the ability of differentiation of SCs reduced rapidly from 11 months old.Conclusion: The number of SCs is related negatively to the age in rats and growing ability of SCs decreased obviously in old rats.
