高血压患者循环内皮祖细胞改变与Ang-1／Tie-2表达的关系研究

目的研究Ang-1／Tie-2在高血压患者外周血中内皮祖细胞的表达及其调控意义。方法选择医院2012年原发性高血压患者30例为观察组，正常体检人群30例为对照组。测定外周血中内皮祖细胞数量和增殖活性，RT—PCR法和Western-blot法检测Ang-1／Tie-2表达。结果观察组内皮祖细胞数量和增殖活性为（6．17±0．89）×103／L和0．784-0．25，低于对照组（6．94±1．12）×103／L和1．14±0．36（P〈0．05）。观察组内皮祖细胞Ang-1和Tie-2的OD值分别为1．65±0．97和4．32±0．84，均低于对照组2．37±1．52和5．02±1．18（P〈0．05）；观察组内皮祖细胞Ang-1和Tie-2的蛋白表达分别为0．06±0．02和0．09±0．04，均低于对照组0．21±0．05和0．20±0．03（P〈0．05）。结论高血压患者内皮祖细胞数量减少，活性减弱，且内皮祖细胞上Ang-1／Tie-2表达下降，提示Ang-1／Tie-2通路的下调可能影响内皮祖细胞的修复功能。

Changes of peripheral blood endothelial progenitor cells related to Ang-1/Tie-2 expression in patients with hypertension

：Objective To study the expression of Ang-1/Tie-2 in peripheral blood endothelial progenitor cells （EPC） in patients with hypertension,and the its role in the regulation of number and function of EPC. Methods Total 30 patients with essential hypertension（ observation group） and 30 health people（ control group） in our hospital in 2012 were enrolled in the study. The amount and proliferation activity of peripheral blood endothelial progenitor cells were measured, the levels of Ang-1/Tie-2 were tested by RT-PCR and Western-blot simultaneously. Results The amount and proliferation ac- tivity of peripheral blood endothelial progenitor cells in the observation group was （6.17 ±0.89） × 103/L and （0.78 ±0.25 ） ,lower than （6.94 ±1.12）×103/L and 1.14 ±0.36 in the control group （ P 〈 0.05 ）. The colorimetric optical densities（OD） of Ang-1 and Tie-2 were 1.65 ± 0.97 and 4.32 ± 0.84 in the observation group, was lower than 2.37± 1.52 and 5.52 ± 1.18 in the control group（P 〈 0.05 ）. The levels of Ang-1 and Tie-2 protein in endothelial progenitor cells were O. 06 ±0.02 and O. 09 ±0. 04 in the observation group,lower than O. 21 ±0.05 and 0.20±0.03 in the control group （ P 〈 0.05 ）. Conclusion The amount and activity of endothelial progenitor cells decreased, while the expression of Ang-1/Tie-2 also down-regulated in patients with hypertension,this promotes that the down-regulation of Ang-1/Tie-2 may probably lead to the changes of endothelial progenitor cells.
