摘    要:目的 研究大鼠黑质区注射脂多糖(LPS)后,纹状体区多巴胺2型受体(D2R)亚型mRNA含量的变化,初步探讨D2R与帕金森病(PD)发病的关系。方法 利用立体定向技术将LPS注人大鼠单侧黑质区,注射后21 d左右,以阿扑吗啡诱导大鼠旋转,RT-PCR方法半定量检测各组大鼠双侧纹状体区D2R两亚型mRNA的表达量。结果 单侧黑质注射LPS的大鼠经阿扑吗啡诱导后向损毁同侧旋转,实验组大鼠纹状体区DL和DS的mRNA表达量损毁侧较对照侧明显增高(P<0.05,P<0.01)。结论LPS损毁大鼠的纹状体部位D2R mRNA含量的变化,可导致大鼠行为的改变,提示D2R可能与PD的发病有一定关系。

Abstract:Objective To study the effect of lipopolysaccharide (LPS) on D2 dopamine receptor ( D2R) subtype mRNA levels in the striatum, and to investigate the relationship between D2R and the pathogenesis of Parkinson's disease ( PD). Methods Lipopolysaccharide was stereostatically injected into the unilateral substantia nigra of rats. Twenty-one days after injection, apomorphine was used to induce rotational behaviour of rats, and at the same time the abundance of both D2S and D2L mRNA was evaluated in the striatum by RT-PCR. Results Compared with the control rats, some rats developed apomorphine-induced rotational behaviour ipsilaterally. There was also marked elevation in the mRNA levels of both isoforms after LPS treatment. Conclusion LPS intranigral injection increased D2L and D2S mRNA levels in the striatum of the injected side, which resulted in the change of behaviour. It suggests that dopamine D,, receptor may be implicated in the pathogenesis of PD.
