弓形虫不同组分抗原免疫保护性的研究

摘    要:目的 探讨弓形虫体不同组分抗原诱导的细胞免疫应答及其对宿主的免疫保护力。 方法 应用SDS PAGE电泳对弓形虫体超声粉碎抗原进行分离、纯化 ,选择特异性抗原组分进行配伍 ,然后与福氏佐剂混合免疫小鼠 ,检测多项免疫指标 ,并观察弓形虫对各免疫鼠的攻击力。 结果 SDS PAGE电泳显示 ,弓形虫体抗原有 7条特异抗原带 ,对其中 4条 (P67、P3 5、P3 0、P2 2 )分离、纯化、配伍后分别免疫小鼠 ,各实验鼠脾脏的CD8+ T淋巴细胞及其脾细胞培养液中IL 2、IFN γ的滴度均随免疫时间延长而增加 ,其中以P3 0 /3 5配伍组增高最明显 ,并且该组合诱导的免疫保护力最强 ,攻击感染后平均存活时间 (7.0 5d)明显长于其它实验组和对照组 (P <0 .0 5 )。 结论 P3 5和P3 0蛋白联合使用能诱导小鼠产生较高的细胞免疫和较强的免疫保护力 ,二者可能成为弓形虫多价亚单位疫苗的侯选抗原。

Abstract:Objective To research the cellular immunity and protective effect induced by the combinations of different proteins from Toxoplasma gondii . Methods The supersonic smashed proteins of T. gondii were seperated and purified by SDS-PAGE, then the specific proteins from the separated and purified proteins were selected as antigens which will immunize mice with the forms of different combinations and several indexes of immunity and the ability of anti- T. gondii infection were detected. Results Seven specific proteins were found and four of them (P67, P35, P30, P22) were selected as antigens and purified. After immunization, the combination of P30 and P35 induced strong protective effect on anti- T. gondii infection. The concentration of CD8 + T lymphocyte, IL-2 and IFN-gamma in mouse spleen induced by the combination of P30 and P35 proteins was the highest among all groups. The mean survival time of mice was 7.05 days after attacked by T. gondii in P30/35 group which was the longest survival time( P <0.05). Conclusion The combination of P30 and P35 can induce strong cellular immunity and offer strong protective effect( P <0.05), so this combination is fit to be used in subunit vaccine of T. gondii .
