摘    要:目的 研究参附注射液对大鼠移植肝脏缺血-再灌注损伤的保护作用及其机制.方法 将雄性SD大鼠分为参附组、对照组和肝脏缺血-再灌注损伤模型组.参附组在获取供肝前经腰静脉灌注参附注射液(2mL/100 g),对照组注入等量0.9%氯化钠注射液,模型组不予处理.光镜观察术后180 min肝脏病理学变化.分别于肝移植完成后0,60,180 min测定大鼠血清丙氨酸氨基转移酶、天门冬酸氨基转移酶和肿瘤坏死因子-α水平.结果 再灌注180 min后,参附组病理损害程度较对照组和模型组轻;再灌注60 min和180 min后,参附组丙氨酸氨基转移酶、天门冬酸氨基转移酶和肿瘤坏死因子-α因子水平明显低于对照组和模型组(P<0.05).结论 参附注射液通过降低炎性因子肿瘤坏死因子-α表达水平,对大鼠移植肝脏缺血-再灌注损伤有明显保护作用.

Abstract:Objective To investigate the protective role and mechanism of Shenfu Injection(SF) in ischemia-reperfusion injury of trans- planted liver in rats. Methods Male SD rats were divided into three groups: SF group,control group and ischemiareperfusion injury (I-RI) group. The SF group was infused SF 2 mL/100 g by lumbar vein before getting donor liver. The control group was infused with equivalent 0.9% sodium chloride injection without any treatment for I- RI group. The level of TNF- α, ALT and AST were de- tected at 0,60,180 min ,after liver transplantation. The histopathological damage degree of grafted liver was observed by light micro- scope. Results The degree of histopathological damage at 180 min after reperfusion in the SF group was lower than that in the control and the I-RI group. The levels of TNF-α,ALT and AST at 60,180 rain after reperfusion were significantly decreased in the SF group than the control and I- RI groups (P 〈 0. 05). Conclusion SF has significant protective effect on transplanted liver in rats via reducing the expression of TNF-α.
