川芎总黄酮提取优化及小鼠体外抗氧化作用

摘    要:目的:研究川芎总黄酮(LCTF)的提取和抗氧化作用。方法:总黄酮的提取采用微波提取和索氏提取2种方法,设置不同的因素水平,从而确定最佳提取方案;抗氧化研究采用总黄酮对血清抗活性氧能力的影响,对小鼠肝匀浆生成MDA的影响,对脂肪氧合酶活性的影响,总黄酮对H2O2诱导小鼠氧化溶血的影响。结果:微波提取总黄酮最佳因素提取频率为530W,提取时间为20min,70%乙醇,液固比为15∶1,得率为1.008%。抗氧化作用表明,2.00g.L-1LCTF对脂肪氧合酶的粗酶活性抑制率超过50%;LCTF在0.5~10.0g.L-1范围内,具有较强的体外抗活性氧能力,并能减少小鼠肝匀浆MDA的生成,可抑制H2O2诱导的小鼠红细胞溶血。结论:微波提取LCTF得率较高,且LCTF具有较强的抗氧化能力,并与总黄酮溶液的浓度具有剂量-效应关系。
