醒脑液对双转基因痴呆小鼠行为学及血浆中胆碱能系统相关酶的影响

摘    要:目的:观察醒脑液早期干预对阿尔茨海默病模型APPswe/PS△E9双转基因小鼠行为学及血浆中胆碱能系统相关酶的影响。方法:将75只PAP双转基因小鼠随机分为空白对照组(模型组)、阳性对照组、用药高剂量组、用药中剂量组和用药低剂量组,每组各15只。15只遗传背景相同的转基因阴性的小鼠作为正常动物组。所试药物稀释至相同体积灌胃给药,正常动物组及空白对照组给以等体积蒸馏水灌胃,连续灌胃4个月。采用Morris水迷宫测定小鼠空间学习记忆能力,避暗箱测定其条件学习记忆能力。使用比色法检测小鼠血浆中乙酰胆碱(Ach)、乙酰胆碱转移酶(ChAT)及乙酰胆碱酯酶(AchE)的活性。结果:与正常组比较,模型组小鼠的学习记忆能力均有所下降,血浆中Ach、AchE的含量均下降。与模型组比较,阳性对照组、用药高剂量组及用药中剂量组小鼠学习记忆能力提高,且血浆中Ach含量均有所提高,而AchE含量则出现下降。结论:醒脑液能改善阿尔茨海默病模型小鼠的学习记忆能力,其作用机制可能与调控胆碱能系统有关。

Abstract:Objective : To observe the effect of Xingnaoye on behavioral science and related enzymes of cholinergic sys- tem in plasma of Alzheimer disease model Appwe/ps A E9 double transgenic mice. Method :75 PAP double transgenic mice were randomly divided into blank control group (model group), positive control group, high dose group, medium dose group and low dose group, with 15 mice in each group. 15 transgenic negative mice of the same genetic background Were as the normal control group. Xingnaoye was diluted to the same volume for three dose groups, and normal control group and blank control group were given an equal volume of distilled water, orally for 4 months. The Morris water maze was used to determine learning and memory ability of mice in space, avoiding black - box to determine one in condition. The colorimetric determinations of acetylcholine (Ach), acetylcholine transferase (CHAT) and acetylcholinesterase (AchE) in plasma for mice were carried out. Result: Compared with the normal group, the learning and memory ability of mice and the contents of Ach and AchE in model group were decreased. Compared with the model group, the learning and memory ability of mice and the content of Ach in positive control group, high dose group and medium dose group were increased, while the content of AchE was decreased. Conclusion : Xingnaoye can improve the learning and memory ability of Alzheimer; disease model mice and the mechanism may be related to regulation of cholinergic system.
