摘 要: | 目的:研究卵母细胞体外成熟(in vitro maturation,IVM)技术中,核抑制剂Roscovitine联合胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)是否协调胞核和胞质的同步成熟。方法:促排周期获取生殖泡期(germinal vesi-cle,GV)卵母细胞164个,分对照组、Roscovitine组、Roscovitine联合IGF-1组3组体外成熟后,其中111个用于研究胞质和胞核是否同步成熟,53个用于卵母细胞内单精子注射(Intracytoplasmic sperm injection,ICSI)判断原核和胚胎发育情况。结果:经过24h培养Roscovitine组胞核成熟率显著低于对照组(P<0.05),但48h累计成熟率各组无显著性差异。体外培养24h或48h将胞核成熟的卵母细胞行皮质颗粒染色以观察胞质成熟度,Roscovitine联合IGF-1组胞质成熟率显著高于其他两组(P<0.05)。对体外培养24h胞核成熟的卵母细胞行ICSI,Roscovitine联合IGF-1组2-细胞胚胎形成率高,胚胎阻滞率低,但无显著性差异。结论:Roscovitine联合IGF-1有利于协调浆核同步成熟。

Study of Roscovitine combined with IGF -1 on oocytes'nuclear and cytoplasm synchronize maturation in vitro |

Abstract: | Objective:To find out whether Roscovitine combined with IGF-1 can do good to oocytes' nuclear and cytoplasm synchronic maturation in vitro.Methods:From controlled ovarian hyper stimulation (COH),164 GV oocytes were got,then divided into control group,Roscovitine group and Roscovitine combined with IGF-1 group.111 oocytes were used for the study of whether nuclear and cytoplasm synchronize during in vitro maturation,and the other 53 oocytes were used for ICSI after maturation to observe the fertilized information.Results:After 24 h cultivation,the group used Roscovitine only had lower maturing rate (P<0.05) compared to the control group.But three groups shown no significant differences at 48 h' cultivation.At 24 h,we stained the cortical granule of oocytes that excluded PB1,cytoplasmic maturing rate was significant higher in the group of Roscovitine combined with IGF-1(P<0.05),but we did not see that at 48h.Some nuclear-matured oocytes were used for ICSI after 24h cultivation,the group of Roscovitine combined with IGF-1 had higher 2-cell embryo formation rate and lower embryo blockage rate,but there was no significant sense.Conclusion:Roscovitine combined with IGF-1 can coordinate oocytes'nuclear and cytoplasm synchronic maturation in vitro. |
