摘    要:
目的:建立以高效液相色谱法同时测定附子中乌头碱、新乌头碱、次乌头碱含量的方法。方法:色谱柱为HypersilC8,流动相为甲醇-0·04mol/L三乙胺(50∶50,磷酸调节pH值至6),流速为1·0ml/min,检测波长为230nm,柱温为40℃。结果:乌头碱、新乌头碱、次乌头碱进样量分别在0·0008μg~0·19μg(r=0·9996)、0·00159μg~0·40μg(r=0·9998)、0·00186μg~0·47μg(r=0·9997)范围内与峰面积积分值线性关系良好,平均回收率分别为94·2%(RSD=2·0%)、95·2%(RSD=1·3%)、96·4%(RSD=1·8%)。结论:本方法简便、准确、分离效果好、线性范围宽、灵敏度高,可用于本品的质量控制。

Abstract:
OBJECTIVE:To establish an HPLC method for simultaneous content determination of aconitine,mesaconitine,and hypaconitine in Fuzi.METHODS:Hypersil C 8 was used as chromatographic column,methanol-0.04mol/L triethylamine (pH adjusted to6with H 3 PO 4 )(50∶50)as mobile phase.The detection wavelength was set at230nm,the flow rate at1.0ml/min and the column temperature at40℃.RESULTS:The sample sizes of aconitine,mesaconitine,hypaconitine had good linear re-lationship with their peak areas in the range of0.0008μg~0.19μg(r=0.9996),0.00159μg~0.40μg(r=0.9998),and0.00186μg~0.47μg(r=0.9997),respectively.Their average recoveries were94.2%(RSD=2.0%),95.2%(RSD=1.3%),and96.4%(RSD=1.8%),respectively.CONCLUSION:The established method is convenient and accurate with satisfactory separation effect,wide linear range,and high sensitivity,and can be available for the quality control of Fuzi.
