BMP2mRNA在骨折愈合过程中的表达及意义

摘    要:目的:探讨骨形成蛋白(BMP)在骨诱导过程中的作用机理.方法:建立骨折愈合模型,利用原位杂交方法检测骨折愈合过程中BMP2mRNA的表达.结果:骨折后第5日间充质细胞、成骨细胞内均见阳性信号,第7日开始信号明显增强,软骨细胞内也可见阳性信号,第14日后信号逐渐减弱.结论:在成骨活动活跃时,间充质细胞、成骨细胞、软骨细胞均有BMP2mRNA的转录,然后编码形成BMP2蛋白来促进及调节骨折的修复.

Abstract:Objective: To elucidate the action of bone morphogenetic protein (BMP) during bone induction. Methods: An experimental model of fracture healing was used to detect BMP2 mRNA by non radioactive ISH. Results: By the 5th day after the fracture, mesenchymal cells and some osteoblasts showed BMP2 positive signals. By day 7, BMP2 mRNA expression was intense, chondrocytes also showed BMP2 positive signals. By day 14, reduced BMP2 mRNA expression was observed. Conclusion: These findings suggest that mesenchymal cells, chondrocytes and ostoblasts may transcribe BMP mRNA, synthesize BMP during active bone formation.
