胆囊癌组织中BRAF和Caspase-3的表达与相关性研究

目的探讨胆囊癌组织中BRAF和Caspase-3的表达水平，了解胆囊癌组织中BRAF和Caspase-3的表达的相关性。方法采用免疫组织化学法检测68例组织中BRAF和Caspase-3的蛋白表达水平（49例胆囊癌、9例胆囊腺瘤、10例胆囊炎）。结果在所检测的胆囊癌组织中，BRAF阳性表达率为65．3％（32，49），明显高于胆囊炎和胆囊腺瘤组（P〈0．01）；而Caspase-3在胆囊癌组织中阳性表达率为42．9％（21，49），明显低于胆囊炎和胆囊腺瘤组（P〈0．01）。BRAF表达与肿瘤的细胞学分级、Nevin分期及性别年龄均无统计学意义（P〉0．05）。Caspase-3表达与分化程度有关而与Nevin分期及性别年龄等无关。BRAF与Caspas-3表达呈负相关（r=-0．373，P=0-008）。结论对BRAF和Caspase-3的联合检测可能作为胆囊癌诊断、预后判断的指标。

Objective To dectect the expression of BRAF and Caspase-3 in gallbladder carcinoma and to explore their correlation with invasion.Methods The expression of BRAF and Caspase-3 were detected with immunohistochemistry in 68 cases(include 49 cases of gallbladder carcinoma and 9 cases of gallbladder aderloma and 10 cases of cholecystitis).Results In response to the detected in gallbladder carcinoma tissue,the positive expression rate of BRAF was 65.3%(32/49),the positive expression rate of BRAF was significantly higher than that of cholecystitis and gallbladder adenoma group.But,Caspase-3 in gallbladder carcinoma tissue positive expression rate of was 42.9%(21/49),the positive expression rate of Caspase-3 was significantly lower in cholecystitis and gallbladder adenoma group.BRAF expression and tumor cytologic grading showed no relationship,while the expression of Caspase-3 was associated with differentiation.The expression of survivin correlated inversely with Caspase-3(r =-0.373,P = 0.008).Conclusion Therefore,the joint detection of BRAF and Caspase-3 expression may serve as an indicator for judging diagnosis and prognosis metastatic potential of gallbladder carcinoma.
