ING1基因蛋白在胃癌组织中的表达及临床意义

摘    要:目的探讨p33ING1基因蛋白与胃癌发生及病理生物学关系及临床意义.方法对71例胃癌、12例胃粘膜不典型增生、18例对照组,应用Envision免疫组化法,检测p33ING1.结果粘膜不典型增生组及对照组中p33ING1均阳性表达,而胃癌中表达62.0%(44/71)有明显下降,与前两组之间有显著差异(P<0.01);胃癌中p33ING1表达与肿瘤的浸润、淋巴结的转移及分化有关(P<0.01).结论p33ING1是抑癌基因,在胃癌中表达下降,对胃癌发生、发展可能起重要作用,p33ING1与p53野生型基因互相协同、依赖,抑制细胞生长、促进凋亡.p33ING1的水平,对p53介导的基因治疗尤为重要,应予检测.

Abstract:Objective:To discuss the pathological relationship between p33 ING1 and stomach cancer and its clinical signification. Method:In 71 cases of stomach cancer, 12 cases of gastric mucous membrane atypical hyperplasia and 18 cases of control group, using Envision immunohistochemical method, we examined the expression level of p33 ING1 in each group. Result:p33 ING1 expression in mucous membrane atypical hyperplasia group and control group is positive,while the expression in stomach cancer group is extremely low(62.0% 44/71), which is obviously different from the other two groups(P<0.01). p33 ING1 expression in stomach cancer is related to the tumor grouth, lymphnode metastasis and tumor polarization (P<0.01). Conclusion:p33 ING1 is a tumor supressor gene and its expression in stomach cancer is low.It may play important roles in the occurrance and development of stomach cancer.p33 ING1 cooperated with p53 wild type suppresses cell growth and accelerates the death of cells. The p33 ING1 level is very important to gene therapy based on p53, and should be examined.
