CpG-ODN调节小鼠免疫细胞对HBsAg体液免疫应答性的初步研究

摘    要:目的:初步探寻能够刺激小鼠免疫系统产生针对HBsAg的体液免疫应答的最佳CpG-ODN序列。方法:应用正交设计法设计含有不同序列的10例CpG-ODN免疫小鼠,以研究影响CpG-ODN免疫活性的6个因素;免疫时间,CpG基序侧翼核苷酸、5‘‘‘‘‘‘‘‘-poly(A)、硫代修饰,回文结构和CpG-基序数量,筛选出量优组合的CpG-ODN。结果:10例CpG-ODN与HBsAg一起免疫小鼠均可不同程度地诱导小鼠抗HBsAg抗体水平的变化,显示出CpG-ODN较强的佐剂效应。结论:CpG-ODN与HBsAg一起免疫小鼠可诱导小鼠产生强烈的抗体应答,以六个因素均存在时CpG-ODN的免疫刺激活性较强,即10号序列5‘‘‘‘‘‘‘‘-AAAAAACGTTAACGTT-3’为本实验中刺激小鼠免疫系统产生针对HBsAg的体液免疫应答的最佳序列。

Abstract:Objective To primarily investigate the optimal CpG ODN sequence which could stimulate the mouse's immune system to produce the humoral immune response to HBsAg.Methods Ten kinds of CpG ODNs were designed which had different sequences by using orthodogonal design method,and immunized the mice with them.By studying the six factors immune time,flanking nucleotide of CpG motif,5' poly(A),phosphorothioate modification,palindrome structure and the amount of CpG motif,which could influence the immune activity of CpG ODN,was the optimal CpG ODN selected.Results Accompanied by HBsAg,all these ten kinds of CpG ODNs could differently change the antibody level against HBsAg in the mice,thus the powerful adjuvant effect of CpG ODNs was showed.Conclusion When the mice were immunized with HBsAg and CpG ODN,the strong antibody response could be induced in them.The CpG ODN with all the six factors had a stronger immunostimulatory activity,i.e,the tenth sequence 5' AAAAAACGTTAACGTT 3' in our trial was the excellent one among those which could stimulate the mouse's immune system to produce humoral immune response to HBsAg.
