紫茎泽兰提取物对小鼠脾细胞凋亡的影响

摘    要:目的 观察紫茎泽兰提取物对小鼠脾细胞凋亡的影响。方法 小鼠分别以0、75、150、300 mg/kg的剂量经口给予紫茎泽兰提取物,连续7 d后,无菌条件下取小鼠脾脏,制备脾细胞悬液,在RPMI 1640培养基中培养24~48 h,流式细胞术观察脾细胞凋亡发生率,光镜下观察脾脏组织病理学变化。结果 紫茎泽兰提取物各剂量组脾细胞早期凋亡率、晚期凋亡率及死亡率都明显增加。脾脏组织病理学检查可见,红白髓界限不清,白髓减少或消失,淋巴小结结构破坏,淋巴细胞数量较少,排列疏松,皮质层变薄。结论 紫茎泽兰提取物可引起脾脏组织病理学明显改变,脾细胞凋亡率明显增加。

Abstract:Objective To observe apoptosis of splenocytes in mice with Eupatorium adenophorum spreng extract. Methods The mice were gavaged Eupatorium adenophorum spreng extract at dose of 75. 0、150. 0、300. 0 g·kg1·d-1 for 7 days. The spleens were excised from mice under aseptic condition,and single-cell suspension of splenocytes was prepared. After incubation in RPMI 1640 medium supplemented for 24 ~ 48 hours,apoptotic rate of splenocytes was measured. The pathological changes in spleen were observed by optical microscopy. Results By optical microscopy,white pulps were atrophied and splenic nodules were destroyed,cell dissolved fragments. A significant increase of a apoptotic rate in Eupatorium adenophorum spreng extract treate groups compared with the control Conclusion Eupatorium adenophorum spreng extract could cause obvious pathological changes and significant increase of a apoptotic rate in the mouse spleen.
