nmhaFGF对肾脏缺血再灌注损伤大鼠不同时间SOD、MDA、BUN、Cr和尿蛋白含量的影响

摘    要:目的:研究非促分裂型人酸性成纤维细胞生长因子(nmhaFGF)对大鼠肾脏缺血再灌注损伤的保护作用。方法:30只大鼠随机分成3组,每组10只。假手术组:切除动物左侧肾脏,暴露右侧肾脏,分离输尿管,不夹闭肾动脉,经舌静脉注射生理盐水1.5ml;模型组:模型制作成功后舌静脉注射生理盐水1.5ml;nmhaFGF组:模型制作成功后经舌静脉注射40μg/kg的nmhaFGF1.5ml。术后4h、8h、16h、24h、48h、72h和7d检测大鼠血液中SOD、MDA、BUN、Cr以及尿液中蛋白含量的变化。结果:模型组16h血液中SOD开始明显下降,MDA明显上升,24hSOD下降至最低点,MDA含量达到最高点;8h血液中BUN和Cr含量明显高于假手术组,24h血液中BUN和Cr含量达到高峰,7d后BUN和Cr下降。模型组尿液中蛋白含量与假手术组相比各时间段均明显升高,24h达到峰值。nmhaFGF组16h和24h血液中SOD、MDA、BUN和Cr与模型组相比显著下降;尿蛋白含量在24h下降最为显著。结论:nmhaFGF具有抗肾脏缺血再灌注损伤自由基的作用和改善肾小球功能的作用,并且该作用呈现一定的时效关系。

Effects of non-mitogenic human acidic fibroblast growth factor on SOD, MDA, BUN and Cr in ischemia-reperfusion rats at different time

Abstract:Objective: To study the effects of non-mitogenic human acidic fibroblast growth factor (nm-haFGF)on the renal acute isehemia-reperfusion injury in rats . Methods : 30 rats were divided into 3 groups randomly. The rat models of renal ischemia-reperfusion injury were established. 5min later, 40μg/kg nmhaFGF was injected by lingual vein. After the operation at the time of 4h,8h, 16h,24h,48h,72h and 7d , the contents of SOD,MDA, BUN,Cr in blood and the content of protein in urine were detected respectively. Results: In the model group,the activity of SOD in blood went down at the time of 16h and decreased to the mimimum value at the time of 24h ,while the content of MDA reached to its peak at the time of 24h. The contents of BUN and Cr in blood increased compared to the sham operation group from the time of 8h. At the time of 24h the contents reached the peak. 7d later, the contents went down. The content of protein in urine in the model group increased obviously compared to the sham operation group at the time of 4h, 8h, 16h,24h,48h,72h and 7d. At the time of 24h the concentration reached its peak. In the nmhaFGF group, the activity of SOD increased dramatically compared to the model group at the time of 24h,while the content of MDA decreased obviously at the time of 24h. The contents of BUN and Cr decreased dramatically at the time of 16h and 24h compared to the model group. The content of urine protein decreased at the time of 16h and 24h. Conclusion: nmhaFDF could resist the effects of free radicals and inprove the renal glomeruli functional impairment of rats induced by ischemia and reperfusion. The effects are in time-dependent manner to some extent.
