摘    要:
采用大鼠肺泡巨噬细胞(AM)体外培养的方法,对铁矿积尘进行毒性研究。染尘浓度为0、0.2、0.6和1.2mg/ml,染尘1、5、20小时后,通过对大鼠AM功能、形态、细胞膜通透性变化以及脂质过氧化、抗氧化等的观察,初步探讨了铁矿尘对AM的毒作用及其机理。结果表明,染尘后AM吞噬酵母菌能力降低;随铁矿尘浓度增高和染尘时间延长,细胞培养液中LDH活性升高,细胞内ACP活性降低,K^+水平降低,说明铁矿

Abstract:
The effects of iron ore dust on cytotoxicity were examined in a cell culture system of rat alveolar macrophages(AM). The dust concentration in culture medium were 0, 0.2, 0. 6, and 1 2mg/ml. After culture for 1,5,and 20 hours,the cytotoxicity and mechanisms of iron ore dust on AM were studied by the observation of function and morphology of AM and the changes of membrane penetration, lipid peroxidation and antioxidation. The results showed that the AM phagocytosis on Saccharomycete cerevise was depressed, the lactate dehydrogenase activity in the culture medium was increase, the acid phosphatase activity was decreased with the increase of dust dose and time, and the content of potassium in the cells was declined, which indicated that iron ore dust induced the changes of AM membrane penetration. With scanning electon microscopy, many broadened folds and protrusions showing "activated" surface after culture for 1 hour at 0. 2mg/ml were observed, and at 1. 2mg/ml the cell surfaces were damaged,folds and protrusions disapeared and small holes in the surface membrane could be seen. The cell surface structure was nearly smooth, in the meantime the lipid peroxidation was inhanced. The findings revealed that the cytotoxicity of iron ore dust was related to the lipid peroxidation.
