摘    要:目的:观察鸵鸟骨转化多相钙磷陶瓷复合骨髓基质细胞后植入裸鼠皮下的成骨性能。方法:获取兔髂骨松质骨骨髓基质细胞,体外分离、扩增、诱导后接种于多相钙磷陶瓷支架。支架/细胞复合物植入裸鼠背部皮下,单纯支架材料植入作为对照。植入后4周、8周取材,通过大体、组织学观察评价成骨活性。结果:支架/细胞复合物植入后4周,新骨主要以软骨为主,部分区域见少量成熟骨,可见软骨内成骨方式。植入后8周,材料表面和孔隙内有更多的成熟骨形成,部分区域有血管和多核巨细胞分布。结论:支架/细胞复合物显示良好的成骨活性,鸵鸟骨转化多相钙磷陶瓷可以用于骨组织工程支架材料。

Ectopic Osteogenesis of Multiphasic Calcium Phosphate Ceramic Transformed by Ostrich Bone Loaded with Bone Marrow Stromal Cells
Abstract:Objective: To observe the osteogenic potential of multiphasic calcium phosphate ceramic transformed by ostrich bone loaded with bone marrow stromal cells( BMSCs) in a nude mice model. Methods: BMSCs were obtained by harvesting the cancellous bone from the iliac crest of a rabbit. Then the BMSCs were isolated , expanded and induced in vitro and seeded in the multiphasic calcium phosphate ceramic scaffolds. The scaffold/BMSCs composites were implanted subcutaneously into the backs of nude mice. Implantation of the multiphasic calcium phosphate ceramic alone was used as control. At 4 and 8 weeks after implantation the specimens were harvested and the osteogenetic activity were evaluated by gross and histologic observation. Results: At 4 weeks after implantation, the newly formed bone were composed mainly by cartilaginous bone while a small part of the mature bone were found in some regions. The endochondral bone formation pattern were found. At 8 weeks after implantation, much more mature bone had formed in the surface and pores of the ceramics than before. Lots of blood vessels and multinucleated cells were seen in some places. Conclusion: The scaffold/BMSCs composites showed good osteogenetic activity. It suggested that the multiphasic calcium phosphate ceramic transformed by ostrich bone could be used as a bone tissue engineering scaffold.
