摘    要:
目的:观察氧化低密度脂蛋白(ox-LDL)对大鼠胸主动脉血管平滑肌细胞(VSMCs)骨保护素(OPG)及核因子-κB受体活化因子配体(RANKL)表达的影响。方法:体外培养大鼠VSMCs细胞株(A7r5),采用不同浓度Ox-LDL(75、150、300μg/mL)或不同时间(4、12、24h)处理构建VSMCs损伤模型,分别采用定量PCR、Western blot方法检测大鼠主动脉OPG、RANKL表达情况。结果:经过oxLDL干预后,VSMCs高表达OPG而低表达RANKL,且呈现浓度和时间的依赖性。结论:ox-LDL能够促进大鼠VSMCs OPG表达,并同时抑制RANKL表达。

Abstract:
Objective: To investigate the effects of oxidized-low density lipoprotein (ox-LDL) on the expression of osteoprotegerin (OPG) and receptor activator of NF-κB ligand (RANKL) in rat aortic vascular smooth muscle cells (VSMCs). Methods: The cultured VSMCs (A7rS) were treated with ox-LDL by different concentration or different period of time, and OPG and RANKL expression in rat thoracic aortas were detected by qRT-PCR and Western blot analysis. Results.. Compared with control group, OPG mRNA and protein levels were increased in a dose and time-dependent manner when VSMCs were induced by ox-LDL, while RANKL mRNA and protein levels were decreased. Conclusion: These findings suggest that ox-LDL induced the expresson of OPG, and suppressed the expression of RANKL in VSMCs.
