蛋白酪氨酸磷酸酶1B与胰岛素的信号转导

摘    要:
蛋白酪氨酸磷酸酶1B(PTP-1B)是一种胰岛素信号转导的负性调节因子,通过使胰岛素受体及其底物等信号蛋白的酪氨酸去磷酸化而阻断胰岛素的信号转导通路,其负调节作用具有组织特异性,多种因素影响其催化活性,另外PTP-1B在胰岛素信号转导通路中也与其他蛋白酪氨酸磷酸酶相互协调作用。由于PTP-1B与2型糖尿病、肥胖症等代谢性疾病的关系密切,开发小分子PTP-1B抑制剂对该类疾病的防治提供了新的思路。

Abstract:
Protein tyrosine phosphatase 1B(PTP-1B)is the negative regulatory factoes in insulin signal transduction,which can inhibit the insulin signal transduction pathway by dephosphorylating the phosphotyrosine of many signal protein such as insulin receptor and its substrate.Its negative regulatory effect has tissue specificity,and many factors can affect its catalytic activity.Besides that,PTP-1B also has intercoordination with other protein-tyrosine-phosphatase in insulin signal transduction pathway.Because of ...
