滑菇多糖的制备工艺

摘    要:
目的确定制备滑菇多糖的最佳工艺路线。方法采用酶解结合水浸提的联合工艺制备滑菇子实体多糖。结果确定浸提比为 1∶1 0 ,纤维素酶 (0 .0 5 %加酶量 ,pH 5 .0 ,45℃ )酶解 60min ,胰蛋白酶 (0 .0 5 %加酶量 ,pH 8.0 ,2 5℃ )再酶解 45min ,之后 80℃保温水浸提 90min为最佳制备路线。结论该工艺制备滑菇多糖的浸提率较单纯的酶解或水浸提分别提高了 38%和 67% ,而且容易纯化 ,是行之有效的制备方法

Abstract:
PurposeTo define the optimum of extracting polysaccharide from Pholiota nameko. Methods Using a united technology of the combination of enzyme decomposing and water extracting methods to obtain the PNP.ResultsThe optimal conditions of extracting PNP were as follows:adding 10 times of water volume as much as that of sample and adding cellulose enzyme 0.05% weight of sample,extracting 60 min at 45℃ (pH 5.0), then adding trypsin of 0.05% weight of sample, extracting 45 min at 25℃ (pH 8.0), at last, water extracting 90 min at 80℃.ConclusionCompared with the simple water extracting and enzyme extracting methods, united treatment made yield of polysaccharide increasing by 38% and 67%, and the polysaccharides were easier to be purified. In a word, the method was feasible.
