摘 要: | 目的探讨链脲佐菌素诱导小鼠实验性糖尿病模型的建立。方法一次性腹腔注射链脲佐菌素(150mg/kg)建立糖尿病小鼠模型,正常小鼠作对照。每周监测其体重和血糖变化,连续观察8周;摘取胰腺和肾脏,常规HE染色,光镜下进行组织形态学观察。结果与对照组相比,模型组小鼠体重明显减轻(P〈0.01),血糖持续升高(P〈0.01);模型组小鼠胰岛形态不规则,体积萎缩变小,胰岛内分泌细胞数量明显减少;肾小球明显肥大,且毛细血管数目显著增多。结论单剂量腹腔注射链脲佐菌素(150mg/kg)诱导糖尿病小鼠模型,是糖尿病相关研究较理想动物模型之一。

Abstract: | Objective To study the establishment of streptozotociminduced experimental diabetic model in mouse. Methods Diabetic mouse model was induced by intraperitonea! inject!on of a single dbse o{ streptozotocin 150 mg/kg. Normal mice served as control. The body weight and blood glucose level of mice were weekly measured at certain time for 8 weeks. The pancreas and kidney of the mice were dissected,and the sections of pancreas and kidney were stained with hematoxylin and eosin for morphological observation under light microscope. Results In comparison with the con- trol mice, the body weight was significantly decreased, and the blood glucose level was significantly increased in diabetic mice (P%0.01). The pancreatic islets of diabetic mice were irregular and shrinked obviously. The endocrine cells of pancreatic islets were markedly decreased in diabetic mice. Otherwise,the enlarged glomeruli and increased blood capil- lary in glomeruli were also observed in diabetic mice. Conclusion The diabetic mouse model induced by intraperitoneal injection of a single close of streptozotocin 150 mg/kg may be one of the relatively suitable animal model for the study associated with diabetes mellitus. |
