尿毒康合剂对腹膜透析液诱导的大鼠腹膜组织学变化及TGF-β1表达的影响

摘    要:目的:本研究主要是探讨我院制剂尿毒康合剂对于腹膜透析液诱导的大鼠腹膜组织学变化及TGF-β1表达的影响。方法:将36只大鼠按随机数字表分为4组,每组9只:正常对照组不予任何干预;生理盐水对照组腹腔注射生理盐水;腹膜透析组腹腔注射20ml 4.25%葡萄糖透析液;腹膜透析合并用药组在腹腔注射4.25%葡萄糖腹膜透析液20ml,每日用灌胃尿毒康合剂20ml/(kg?d)。疗程4周。4周后,测定腹膜超滤功能和腹膜组织厚度,免疫组织化学方法测定TGF-β1的表达,二硫代二硝酸苯甲酸法显色法和黄嘌呤氧化酶法测定血清及透出液中谷胱甘肽(GSH)和超氧化物岐化酶(SOD)水平。结果:①腹膜透析液组、腹膜透析液合并用药组与正常对照组、生理盐水组相比较,超滤量均显著下降、腹膜组织厚度增加(P<0.05);腹膜透析组与腹膜透析合并用药组比较,超滤量明显减少、腹膜组织厚度增加(P<0.05);②腹膜透析组的大鼠腹膜组织 TGF-β1表达明显增加(P<0.05),腹膜透析合并用药组 TGF-β1表达较腹膜透析组降低(P<0.05);③腹膜透析液组、腹膜透析液合并用药组与正常对照组、生理盐水组相比较,血清及腹透液中GSH和SOD水平水平降低(P<0.05);腹膜透析合并用药组与腹膜透析组比较,血清及腹透液中GSH和SOD水平水平升高(P<0.05)。结论:尿毒康合剂可能具有减轻氧化应激反应和抑制TGF-β1的表达的作用,从而抑制腹膜间皮细胞损伤和腹膜致密层的增厚,达到保护腹膜、延缓腹膜纤维化的作用。

The effect of the Niaodukang mixture on peritoneal dialysis fluid-induced rat peritoneal histological changes and TGF-β1 expression
Abstract:Objective: To investigate the effect of the Niaodukang mixture on peritoneal dialysis fluid-induced rat peritoneal histological changes and TGF-β1 expression. Methods: 36 rats were randomly divided into four groups, 9 in each group. Normal control group was given no intervention, saline control group was given intraperitoneal injection of saline, peritoneal dialysis group were injected 20mL4.25% glucose dialysate, peritoneal dialysis combined with the Niaodukang mixture group was given intraperitoneal injection of 20mL4.25% glucose dialysate and the Niaodukang mixture 20ml/(kg d) every day. The course of treatment was 4 weeks. After 4 weeks, to determine the peritoneal ultrafiltration features and peritoneal tissue thickness, the expression of TGF-β1, the levels of serum and dialysate glutathione (GSH) and superoxide dismutase (SOD). Results: The peritoneal dialysis group and peritoneal dialysis combined with the Niaodukang mixture group were compared with normal control group and saline control group that the ultrafiltration volume significantly decreased, the peritoneal tissue thickness increased (P0.05), the peritoneal dialysis group compared with peritoneal dialysis combined with the Niaodukang mixture group, the ultrafiltration volume significantly reduced, the peritoneal tissue thickness increased (P0.05). Rat peritoneal tissue TGF-β1 expression increased significantly (P0.05) in peritoneal dialysis group, TGF-β1 expression in peritoneal dialysis combined with the Niaodukang mixture group was significantly lower (P0.05) than peritoneal dialysis group. The peritoneal dialysis group and peritoneal dialysis combined with the Niaodukang mixture group were compared with normal control group and saline control group that the level of SOD and GSH was decreased (P0.05) in serum and dialysate, the peritoneal dialysis group compared with peritoneal dialysis combined with the Niaodukang mixture group that he level of SOD and GSH was increased (P0.05) in serum and dialysate. Conclusion: The Niaodukang mixture may reduce oxidative stress and inhibit the expression of TGF-β1, to inhibit peritoneal mesothelial cell injury and peritoneal thickening of the dense layer to protect the peritoneum, delaying the role of peritoneal fibrosis.
