摘    要:目的 研究Abil在肝细胞癌(HCC)组织中的表达情况及其临床意义.方法 采用RT-PCR法检测40例HCC组织及相应癌旁肝组织标本中Abil mRNA的表达情况;免疫组织化学法检测加例HCC组织及相应癌旁肝组织标本中Abil蛋白的表达情况;结合临床病理及随访资料分析Abil表达水平与HCC侵袭转移及预后的关系.结果 Abil mRNA在HCC组织中的表达水平高于相应的癌旁组织(1.32±0.75比0.74±0.52,P<0.05);Abil mRNA在结节性肝癌组织中的表达水平亦明显高于孤立性大肝癌组织(1.72±0.83比1.14±0.57,P<0.05).Abil蛋白主要表达于HCC细胞的胞质内,其表达水平与肿瘤结节数目、是否有包膜、静脉癌栓及Edmondson-Steiner分级密切相关(P<0.05),而与HCC患者的性别、有无肝硬化及肿瘤直径无明显相关性(P>0.05).Abil蛋白高表达组HCC患者的复发转移率明显高于低表达组患者(P<0.05);生存率则明显低于低表达组患者(P<0.02).结论 Abil在HCC组织中的表达水平较癌旁组织高,其表达水平与HCC的结节数目、是否有包膜、静脉癌栓、Edmondson-Steiner分级及临床预后密切相关.

Abstract:Objective To investigate the expression of Abil in hepatocellular carcinama (HCC)and the correlation between its expression level and clinical pathological characteristies as well as prognosis of HCC.Methods Abil expression was determined at both mRNA and protein levels in 40 HCC tissues and their corresponding paracancinomatous liver tissues by RT-PCR and immunohistochemistry.The correlations between Abil expression levels and pathological characteristics as well as prognosis were also analyzed.Results The expression level of Abil mRNA in HCC tissues were significantly higher than those in corresponding paracarcinomatous liver tissue (P<0.05),and the expression level of Abil mRNA in nodular HCC tissues were also significantly higher than those in solitary large HCC tissues.Immunohistochemistry results showed that Abil protein located in cytoplasm of HCC cells and the expression level of Abil protein were significant positive correlated with the number of HCC,capsular formation,venous invasion and Edmondson-Steiner grade (P<0.05).Combined with follow-up data,the results also showed that HCC patients with high Abil protein expression had a higher risk of invasion/metastasis and a shorter survival than those with low Abil protein expression (P<0.05).Conclusions Expression level of Abil is up-regulated in HCC tissues compared with corresponding paracarcinomatous liver tissue and the expression level of Abil is significantly correlated with the number of tumor,capsular formation,venous invasion,Edmondson-Steiner grade and prognosis of HCC.
