摘    要:目的 研究RAB5A基因转染后对肺腺癌侵袭能力的影响。方法 采用重组基底膜侵袭实验 ,观察转染前后的细胞系对基底膜侵袭能力的改变。结果 转染pcDNA3.1 RAB5A正义表达载体的AGZY83 a细胞系基底膜侵袭能力增强 ,转染pcDNA3 AntiRAB5A反义RNA重组质粒的Anip973细胞系侵袭能力明显减弱。结论 RAB5A在人非小细胞肺癌的侵袭及转移特性的形成中具有重要的作用 ,RAB5A的反义分子能够有效地阻断该基因表达的翻译过程 ,在细胞内发挥预期作用。

Abstract:Objective To investigate the effect of RAB5A gene on forming of tumor metastatic nature.Methods Using reconstituted basement membrane invasion technique.We observed the different effect between transfected and untransfected cell lines on basement membrane invisive ability.Results The invasiveness of AGZY83 a cells of transfected pcDNA3.1 RAB5A was higher than parent line, and Anip973 cells of transfected pcDNA3 AntiRAB5A was lower than Anip973 cells. Conclusion The over expression of RAB5A gene plays important role in the formation of tumor invasion phenotype and metastatic nature of human lung adenocarcinoma cells. Antisense RNA can interrupt the translation of RAB5A, and exert the expect effects.
