大豆异黄酮和叶酸对同型半胱氨酸糖尿病动物模型免疫功能的影响

摘    要:目的研究大豆异黄酮和叶酸对同型半胱氨酸(HCY)糖尿病模型动物免疫功能的影响。方法雄性健康昆明小鼠32只,随机分为正常对照组(NS)、高同型半胱氨酸糖尿病组(HCYDM)、大豆异黄酮组(SIF)、大豆异黄酮加叶酸处理组(SIFFA)各8只。通过尾静脉注射链脲佐菌素(STZ)和腹腔注射HCY复制HCY糖尿病动物模型,SIF组及SIFFA组给予大豆异黄酮和叶酸进行干预。检测小鼠脏器/体质量比值、血清溶血素、腹腔巨噬细胞吞噬功能、小鼠碳粒廓清试验。结果与NS相比较,HCYDM组的巨噬细胞吞噬率及吞噬指数、校正碳廓清指数、血清溶血素值、碳廓清指数、脾脏指数和胸腺指数等均显著降低(P<0.05,P<0.01);与HCYDM组相比较,SIF组和SIFFA组巨噬细胞吞噬率、吞噬指数和胸腺指数显著增强(P<0.05,P<0.01);SIFFA组校正碳廓清指数和脾脏指数显著增加(P<0.05),SIF组增加不显著(P>0.05);与SIF组比较,SIFFA组脾脏指数显著升高(P<0.01)。结论大豆异黄酮和叶酸有协同作用,可提高DM模型鼠脾脏和胸腺指数,促进巨噬细胞的吞噬能力,有较好的免疫恢复功能。

Abstract:Objective To research the effect of soybean isoflavone and folic acid on immune function of mice models with homocysteine diabetes.Methods Thirty-two male KunMing mice were randomly divided into 4 groups,which included normal control group(NS),homocysteine diabetes group (HCYDM),soybean isoflavone group(SIF),soybean isoflavone and folacin group(SIFFA).HCY diabetes models were made by vena caudalis injection of streptozocin and intraperitoneal injection of HCY.SIF group were interfered by infection of soybean isoflavone.SIFFA group were interfered by infection of soybean isoflavone and folacin.Detecting the ratio of organ to weight,serum hemolysin,peritoneal macrophage phagocytosis,mice carbon clearance test.Results Compared with NS group,the serum hemolysin,mice carbon clearance test and the index of thymus and spleen decreased,and peritoneal macrophage phagocytosis decreased(P<0.05,P<0.01) in HCYDM group.Compared with HCYDM group,the rate and index of peritoneal macrophage phagocytosis and the index of thymus were significantly increased in SIFFA and SIF group(P<0.05,P<0.01) and the index of carbon clearance and spleen were significantly increased in SIFFA group(P<0.05),but were not significantly increased in SIF group(P>0.05).(3)Compared with SIF group,the spleen index were significantly increased(P<0.01)in SIFFA group.Conclusion Soybean isoflavone and folic acid can improve diabetic mice spleen and thymus organ coefficient,enhanced macrophage phagocytosis,increase the production and release of serum recover immune function.
