摘    要:目的 本研究通过神经再生室对表皮生长因子受体酪氨酸酶抑制剂在面神经损伤后再生过程中的作用,旨在观察局部应用表皮生长因子受体酪氨酸酶抑制剂对面神经再生作用的影响,并为临床治疗面神经损伤提供治疗方法.方法 将新西兰兔20只随机分成两组,切断双侧面神经上颊支制成面神经再生室模型,A组在再生室内注入生理盐水0.1ml;B组在再生室内注入表皮生长因子受体酪氨酸酶抑制剂0.1ml.观察神经损伤局部免疫反应、组织形态学及图像分析、电生理学.结果 1周时B组面神经损伤局部的淋巴细胞浸润程度较A组轻.4、6周时实验组的神经传导速度和组织形态学检查均优于对照组.结论 局部应用表皮生长因子受体酪氨酸酶抑制剂具有神经营养作用,可加速神经功能恢复,促进神经生长.

Abstract:Objective To evaluate the effect of epidermal growth factor receptor-tyrosine kinase inhibitor(EGFR-TKI) on expediting nerve regeneration of rabbit facial nerve in regeneration chamber and to look for a proper way to treat peripheral nerve injuries.Methods Twenty rabbits were randomizely divided into 2 groups received a neurotomy to bilateral facial nerves, then the broken nerves were reconnected with silicon tube to make regeneration chamber. The regeneration chamber was filled with either normal saline(group A) or 10ng/ml EGFR-TKI (group B).Local immunoreaction , histological changes and electrophysiology were observed at designative time after neurotomy.Results At 1 week postoperation the extent of local lymphocytes infiltration in group B were less than that in group A .All results in group B were better than that of group A at 4 weeks and 6 weeks.Conclusion Local administration of 10ng/ml EGFR-TKI can facilitate nerve regeneration and promote functional recovery.
