高血压大鼠模型主动脉僵硬度的超声多普勒评价

摘    要:目的建立一种大鼠模型超声多普勒颈-股脉搏波传播速度(PWV)准确测量方法,将其应用于自发性高血压大鼠(SHR)及其对照组中,验证该方法能否区分SHR已知增加的动脉僵硬度(AS),并探讨其应用价值。方法纳入雄性SHR大鼠12只,周龄、体质量相当的健康对照组Wistar Kyoto(WKY)大鼠12只。大鼠颈-股PWV的测量采用超声多普勒法完成。结果 SHR与WKY大鼠在周龄、体长、体质量、测量期间的心率等指标对比的差异均无统计学意义。SHR大鼠的收缩压、舒张压显著高于WKY大鼠(176 ± 4)mmHg vs (121 ± 5) mmHg,(123 ± 4) mmHg vs (85 ± 5) mmHg,均P < 0.01];SHR大鼠的颈-股PWV测值与对照组相比显著增高(6.5 ± 1.0)m/s vs (4.5 ± 0.9) m/s, P < 0.01];Bland-Altman一致性检验显示两组大鼠的颈-股PWV测值在观察者间具有良好的重复性。结论超声多普勒颈-股PWV测量方法能够精确测量SHR大鼠模型脉搏波传播时间和距离,藉以评价其AS。基于超声多普勒的PWV测量是一种评价大鼠模型AS的可靠方法。

Abstract: AIM To explore a method for measuring carotid-femoral Pulse Wave Velocity (PWV) by Doppler ultrasonography in rat models and apply it in an aortic stiffness evaluation in spontaneous hypertension rats (SHR) compared to a control group. To test whether this method can differentiate increased arterial stiffness (AS) of SHR and its application value. METHODS Twelve male SHR and 12 healthy Wistar Kyoto (WKY) rats with the same age and comparable weight were included in the investigation. The carotid-femoral PWV was measured in the two groups by Doppler ultrasonography. RESUTLS There were no statistically significant differences between SHR and WKY rats in age, body length, body weight, heart rate. The systolic and diastolic blood pressures of SHR were significantly higher than those of control group (176 ± 4) vs (121 ± 5) mmHg, (123 ± 4) vs (85 ± 5) mmHg, P < 0.01]. Compared with the control group, the carotid-femoral PWV values of the SHR were significantly increased (6.5 ± 1.0) vs (4.5 ± 0.9) m/s, P < 0.01]. A Bland-Altman plot revealed good inter-observer reproducibility. CONCLUSION This study presents the method of noninvasively measuring carotid-femoral PWV in a rat models, which could accurately measure the pulse wave transit time and distance. The carotid-femoral PWV measurements differentiated increased arterial stiffness (AS) of SHR from controls. This study may provide a reliable method for noninvasive AS evaluation in rat models in research settings.
