血必净对地塞米松诱导的T淋巴细胞凋亡及凋亡相关基因mRNA水平的影响

目的 观察血必净对地塞米松诱导的T淋巴细胞凋亡及凋亡相关基因mRNA水平的影响.方法 提取BAB/C小鼠睥脏T淋巴细胞并培养,观察血必净对T细胞增殖活性影响的MTT检测;以地塞米松诱导T细胞凋亡,流式细胞仪检测细胞凋亡率,RT-PCR检测Bax、Bcl-2、IL-2 mRNA表达水平,观察血必净对各项指标的影响.结果 血必净可明显提高T淋巴细胞的增殖活性;地塞米松诱导后T淋巴细胞凋亡率增加,Bax mRNA表达升高,Bcl-2、IL-2 mRNA表达下降.血必净干预后可降低T淋巴细胞凋亡,降低Bax mRNA表达,提高IL-2 mRNA表达.结论 血必净可通过降低促凋亡基因表达,提高IL-2表达而降低地塞米松诱导的T淋巴细胞凋亡.

Influence of Xuebijing on apoptosis and apoptotic related gene mRNA levels in T lymphocyte induced by dexamethasone

Objective To observe the influence of Xuebijing on apoptosis and apoptotic related germ mRNA levels in T lymphocyte induced by dexamethasone.Methods The T lymphocyte was obtained from the spleens of BAB/C mice and cultured with Xuebijing.The MTT was measured to assessed the proliferation activity.Apoptosis induced by dexamethasone in T lymphocyte was investigated by flow cytometry.RT-PCR was used to detect the expression of Bax, Bci -2, IL -2 mRNA.Results Xuebijing can improve the proliferation activity of T lymphocyte.Dexamethasone induced the apoptosis of T lymphocyte by increase the express of Bax mRNA and decrease the express of Bcl -2 and IL -2 mRNA.Xuebijing can reduced the apoptotic rations and make the expression of Bax mRNA decreased and IL -2 mRNA improved.Conclusion Xuebijing can ameliorate the apoptosis in T lymphocyte induced by dexamethasone through reducing the expression of pro -apoptotic germ and lifting the expression of IL -2.
