血清中的维生素C含量的高效液相色谱测定法

摘    要:目的建立人血清中维生素C的高效液相色谱(HPLC)检测方法。方法用偏磷酸提取样品中维生素C,应用HPLC分离,PDA二级管阵列检测器检测的定性定量分析。流动相为0.1%磷酸+甲醇=95+5,流速为1.0ml/min,检测波长为243nm。结果在上述色谱条件下,血清中的维生素C能良好的分离,维生素C在1~25μg/ml间呈线性相关,Y=53295.31X-30545.65,r=0.9998;方法的平均相对标准偏差为1.70%,回收率为99.95%;最低检测限为0.05μg/ml。结论该方法可快速、准确、高效地检测血清中维生素C含量。

Abstract:[Objective]To establish the determination method of serum vitamin C by high performance liquid chromatography ( HPLC) .[Methods]Vitamin C was extracted by Metaphosphoric acid,separated by HPLC,and qualitatively and quantitatively analyzed by PDA Diode-array detector. The mobile phase was 0. 1% phosphoric acid + carbinol = 95 + 5,the flow rate was 1. 0 ml/min,the UV detection wavelength was 243 nm.[Results]In the above-mentioned chromatographic condition,serum Vitamin C could be well separated,and Vitamin C showed linear correlation in 1 ~ 25 μg/ml,Y = 53295. 31X-30545. 65,r = 0. 9998; the relative standard deviation is 1. 70% ,recovery was 99. 95% ,and the lowest detection limit was 0. 05μg/ml.[Conclusion]This method can detect serum Vitamin C content rapidly,accurately and efficiently.
