摘    要:顺铂ip或体外直接与肾薄片孵育,均可导致大鼠肾皮质中ATP浓度降低,对氨基马尿酸蓄积减少,丙二醛和血尿素氮升高。大鼠肾动脉结扎1h,放开灌流3h,诱发了类似的肾损害。血管紧张素转化酶抑制剂RamiPriliP给予接受顺铂处理或接受肾动脉结扎的大鼠,或加入到含顺铂的孵育液中,均能明显抑制顺铂和肾缺血再灌注诱发的ATP减少,血尿素氮和丙二醛升高,但对对氨基马尿酸蓄积的减少无保护作用。RamiPril对上述肾损害的保护作用机制可能与其抗脂质过氧化作用有关。

Abstract:The protection of Ramipril (RAM),An angiotensin converting enzyme inhibitor(ACET),against renal injuries induced by cisplatin(DDP),or kidney ischemia-reperfusion in vitro and in vivo was investigated in rats.The study in vitro was performed by using incubation of renal ocrtical slices. Rat kidney ischemia for 1h followed by reperfusion for 3 h,or rats with treatment of DDP(7.5 mg·kg-1,ip) for 48 h,or renal slices incubated for 2 hin DDP-containing(1 mmol,L-1) media·induced decrease of ATP. decrease of accumulation of P-aminohippurate(PAH), rise of malondialdehyde(MDA)in redal cortex and rise of blood urea nitrogen(BUN).RAM(10 mg· kg-1·d-1×2,ip) to rats with treatment of DDP,adding Ramiprilat (active form of RAM) at concentration of 50μmol·L-1 to DDP-containing media in incubation of renal slices,or pretreatment of RAM(10 mg·kg-1,ip)at 24 and 3 h befor occlusion of left renal artery,significantly blunted decrease of ATP as well as the rise of BUN and MDA,but could not prevent decrease of accumulation of PAH in three pathologlc models mentioned above.It Is concluted that RAM can Partially protect renal injuries induced by both DDP and kidney ischemia-reperfusion;the mechanism of protection may be ascribed partially to antilipid peroxidation of RAM.
