p16和nm23-H1蛋白在大肠癌患者中表达的临床意义

摘    要:目的 探讨p16和nm23 -H1在大肠癌患者大肠粘膜组织中的表达及其与预后的关系。方法 应用免疫组织化学标记链霉菌抗生物素 -生物素方法 ,对73例大肠癌患者癌组织中P16和nm23-H1蛋白的表达水平进行了检测。结果 p16和nm23 -H1在大肠癌组织中的表达阳性率分别为32.8 %和43.8% ;分化程度高的癌组织p16的表达阳性率高于分化程度低的癌组织 (P<0.01) ;P16和nm23-H1在无淋巴结转移患者的癌组织中阳性率高于有转移者 (P<0.05、P<0.01)。p16蛋白表达阳性者3年生存率为62.5 % ,而阴性者为32.7 % ;nm23-H1蛋白表达阳性者3年生存率为65.6% ,而阴性者为26.8 % ,阳性表达者与阴性表达者生存率差别有显著性意义 (P<0.05)。p16与nm23-H1蛋白的表达阳性率呈正相关 (r=0.416,P<0.05)。结论 p16和nm23 -H1与大肠癌的发生、发展过程有关 ,可作为预测肿瘤预后的指标。

Abstract:Objective To study the relationship between the expression of p16 and nm23-H1 and prognosis in the patients with colorectal cancer.Methods p16 and nm23-H1 protein in cancer tissue of 73 patients were observed with labeled streptavidin biotin (LSAB) immunohistochemical method.Results The total positive rate of p16 and nm23-H1 was 32.8% and 43.8% respectively in colorectal cancer tissues. The positive expression rate of p16 was significantly higher in well-differentiated cancer than in poorly-differentiated cancer(P<0.05). The positive expression rate of p16 and nm23-H1 was higher in patients with lymph node metastasis than in those without lymph node metastasis(P<0.05,P<0.01).The 3-year survival rate was higher in positive cases of p16 or nm23-H1 protein expression(62.5%,65.6%)than in negative ones(32.7%,26.8%)(P<0.05).There was a significant positive correlation between p16 and nm23-H1 protein expression (r=0.416,P<0.05). Conclusion p16 and nm23-H1 may play a role in the development of colorectal cancer. Expression of p16 and nm23-H1 may serve as a marker for the prognosis of colorectal cancer patients.
