摘    要:目的:研究N-三甲基壳聚糖(TMC)对HaCat细胞细胞膜电位的影响及其与季胺化程度的关系。方法:合成季胺化程度分别为20%,40%及60%的TMC(即TMC20、TMC40、TMC60),用流式细胞仪检测其对HaCaT细胞膜电位的影响。结果:与空白溶剂组比较,TMC能显著降低细胞膜电位,其中,TMC20作用最明显,TMC40组作用最微弱。结论:TMC可降低细胞膜电位,并与其季胺化程度密切相关,这可能是其发挥透皮吸收促进作用的机制之一。

Abstract:Objective: To Study the effect of N-trimethyl chitosan (TMC) on HaCat cell membrane potential and the relationship with its degree of quaternization (DQ). Method: TMCs with DQ of 20%, 40% and 60% (TMC20, TMC40, and TMC60 ) were synthesized. Membrane potentials of HaCaT cells were measured by flow cytometer. Result: Compared with the blank solvent, TMCs could decreased HaCaT cell membrane potential significantly, and the effect of TMC20 was the most significant while that of TMC40 was the mildest. Conclusion: TMCs could decrease the cell membrane potential and the effect had close relationship with their DQ.
