摘    要:目的:研究慢性间歇低氧对幼鼠学习记忆脑区JNK表达的影响。方法:SPF级健康雄性SD幼鼠(3~4周龄)40只,随机分为4组:间歇低氧2周组(2IH组)、间歇低氧4周组(4IH组)、对照2周组(2C组)和对照4周组(4C组)。建立慢性间歇低氧幼鼠模型,以RT-PCR法和Western blot法分别测幼鼠海马及前额叶皮层JNK1 mRNA和磷酸化JNK(p-JNK)蛋白的表达。结果:2IH、4IH组幼鼠海马、前额叶皮层的JNK1 mRNA和磷酸化JNK(p-JNK)蛋白均明显高于相应对照组(P〈0.05或P〈0.01)。结论:JNK的激活可能参与了慢性间歇低氧幼鼠脑损伤过程。

Abstract:Objective:To study the effect of chronic intermittent hypoxia on JNK in memory related cerebral tissues of weanling rats.Methods:Fourty male Sprague-Dawley rats(3-week-old~ 4-week-old) were randomly divided into four groups:2-weeek-CIH group(2IH)4,-week-CIH group(4IH),2-week-control group(2C) and 4-weeek-control group(4C).Intermittent hypoxia model was established with an intermittent hypoxia cabin.The expression of JNK1 mRNA and the phosphorylation levels of JNK(p-JNK protein)in the hippocampus and prefrontal cortex were measured by RT-PCR or Westernblot,respectively.Results:The expression of JNK1mRNA and p-JNK protein in the hippocampus and prefrontal cortex in 2IH and 4IH groups were respectively higher than that in 2C and 4C groups.(P 〈0.05,respectively).Conclusion:The activation of JNK may be involved in the development of injury of cerebral tissues in chronic intermittent hypoxia weanling rats.
