HPLC法同时测定感冒止咳颗粒中绿原酸、葛根素和黄芩苷的含量

目的:建立同时测定感冒止咳颗粒中绿原酸、葛根素和黄芩苷含量的方法。方法:采用高效液相色谱法。色谱柱为Eclipse XDB-C18,流动相为甲醇-ph2.6磷酸溶液(梯度洗脱),流速为0.8 ml/min,检测波长为327 nm(0⁹ min)、250 nm(9.19 min)、250 nm(9.1¹5min)、280 nm(15.115min)、280 nm(15.1³0 min),柱温为25℃,进样量为10μl。结果:绿原酸、葛根素和黄芩苷的检测质量浓度分别在3.730 min),柱温为25℃,进样量为10μl。结果:绿原酸、葛根素和黄芩苷的检测质量浓度分别在3.7³7.5,7.837.5,7.8⁷8.4,15.678.4,15.6¹55.8μg/ml范围内与各自峰面积积分值呈良好的线性关系(r=0.999 9);精密度、稳定性、重复性试验的RSD≤3.41%;平均加样回收率分别为98.9%、100.6%、101.6%,RSD分别为1.51%、1.34%、2.69%(n=9)。结论:该方法简便、快速、重复性好,可用于感冒止咳颗粒的质量控制。

Simultaneous Determination of Chlorogenic Acid,Puerarin and Baicalin in Ganmao Zhike Granules by HPLC

OBJECTIVE： To establish a method for simultaneous determination of chlorogenic acid, puerarin and baicalin in Ganmao zhike granules. METHODS： HPLC method was adopted. The determination was performed on Eclipse XDB-C18 column with mobile phase consisted of methanol-pH2.6 phosphoric acid （gradient elution） at the flow rate of 0.8 ml/min. The detective wavelength was set at 327nm （0-9 min）, 250 nm （9-15 rain）, 280 nm（15-30 min） ; the column temperature was at 25 ℃. The sample size was 10 μl. RESULTS： The linear ranges were 3.7-37.5 μg/ml for chlorogenic acid （r=0.999 9）, 7.8-78.4 μg/ml for pu- erarin （r=0.999 9） and 15.6-155.8 μg/ml for baicalin （r=0.999 9）. RSDs of precision, stability and reproducibility tests were low- er than 3.41%. The average recoveries were 98.9% （RSD=1.51%, n=9）, 100.6% （RSD=1.34%, n=9） and 101.6% （RSD= 2.69%, n=9）, respectively. CONCLUSIONS： The method is simple, rapid and repeatable, which can be used for the quality con- trol of Ganmao zhike granules.
