摘    要:
目的:探讨长链非编码RNA POU5F1B在食管癌细胞中的表达及临床意义。方法:采用实时定量PCR法比较POU5F1B在食管癌患者血浆和正常人血浆中的表达,检测POU5F1B在细胞系中的表达。敲除POU5F1B后,采用CCK-8和克隆形成试验评价沉默POU5F1B对细胞增殖的影响。采用Transwell和划痕实验检测POU5F1B对食管癌细胞侵袭和迁移的影响。结果:本研究中,我们发现POU5F1B在食管癌患者血浆中和细胞系中高表达。si-POU5F1B在体外能显著降低细胞增殖、侵袭和迁移能力。结论:本研究表明POU5F1B在食管癌中充当癌基因作用,为抗食管癌治疗提供了新的策略。

Abstract:
Objective:To investigate the expression and clinical significance of long non-coding RNA in human esophageal carcinoma cells.Methods:The PCR method was used to compare the expression of POU5F1B in EC plasma and normal human plasma.After the knockout of POU5F1B,CCK-8 and clonal formation experiments were used to evaluate the effect of silent POU5F1B on cell line proliferation.The effects of POU5F1B on invasion and migration of EC cell lines were detected by Transwell and scratch test.Results:In this study,we found that POU5F1B was highly expressed in plasma and cell lines of esophageal cancer patients.Si-POU5F1B significantly reduced cell proliferation,invasion and migration in vitro.Conclusion:POU5F1B acts as an oncogene in esophageal cancer and provindes a new strategy for the treatment of esophageal cancer.
