科罗索酸对白血病K562细胞增殖和凋亡的影响

摘    要:目的:探究科罗索酸(corosolic acid)对白血病K562细胞增殖和凋亡的影响。方法:使用不同浓度的科罗索酸处理K562细胞,CCK-8法检测对细胞增殖的影响,流式细胞术检测对细胞周期和凋亡的影响,分光光度法检测对Caspase-3/7,-8,-9活性的影响。结果:科罗索酸能显著抑制白血病K562细胞的增殖,并且这种抑制作用随着科罗索酸浓度升高和培养时间延长而显著增强。科罗索酸处理后的K562细胞中,处于G0/G1期的细胞比率明显升高(P<0.05),S期和G2/M期的细胞明显减少(P<0.05),凋亡细胞明显增多(P<0.05),Caspase-3/7,-8和-9的活性明显增强。结论:科罗索酸能抑制白血病K562细胞的增殖,促进其凋亡,这一效应可能与Caspase活性的增强有关。

Abstract:Objective:To investigate the effect of corosolic acid on proliferation and apoptosis of leukemia K562 cells.Methods:K562 cells were treated with different concentrations of corosolic acid.CCK-8 assay was used to detect the effects on cell proliferation.Flow cytometry was used to detect the effects on cell cycle and apoptosis,and spectrophotometry was used to detect the effects on the activities of caspase-3/7,-8,-9.Results:Corosolic acid significantly inhibited the proliferation of leukemia K562 cells,and the inhibition was both dose-dependent and time-dependent.In K562 cells treated with corosolic acid,more cells were in G0/G1 phase (P<0.05),less cells were in S phase and G2/M phase (P<0.05).The number of apoptotic cells and the activity of caspase-3/7,-8 and-9 also increased significantly after the treatment of corosolic acid (P<0.05).Conclusion:Corosolic acid can inhibit the proliferation and promote apoptosis of leukemia K562 cells,which may be related to the enhancement of caspase activity.
