Effects of Sodium Glutamate and High Glucose Diet on Metabolic Disorders and Memory Impairment in Rats with Type 2 Diabetes

In this study,the authors’ aimed to develop an experimental animal model of type 2 diabetes mellitus( T2 DM) with MSG and HSD,and to explore the effects of sodium glutamate and high-sugar diet on vascular dysfunction and memory impairment in rats with type 2 diabetes. The young male Wistar rats( 20 ~ 30 g) were injected with MSG( 2 or 4 mg/g,intraperitoneal injection for4 days).The rats were also fed with HSD,while the control group was fed with starch diet( SFD) for 150 days.The parameters for regular evaluation were body weight,feed intake,blood glucose level and oral glucose tolerance test( OGTT),lipid distribution,liver and kidney function test,skeletal muscle glucose intake,cognitive function test and microvascular changes using isolated rat aorta.Hematoxylin and eosin staining were used to evaluate the histological changes of kidney. The combination of MSG and HSD could significantly increase the body weight and produce hyperglycemia,dyslipidemia and hyperinsulinemia. Animals developed Frank complications of diabetes,including insulin resistance in skeletal muscles,high blood pressure,vascular dysfunction,kidney disease and dementia.Histological studies have showed that the number of glomeruli in the kidney decrease and the islets of Langerhans show severe hypertrophy and proliferation.The combination of MSG and HSD could successfully induce type 2 diabetes and some diabetic complications.
