黄芩苷对人鼻咽癌CNE-2Z细胞生长的抑制作用

目的 研究黄芩苷对人鼻咽癌CNE-2Z细胞的抑制作用及可能机制。方法 用倒置显微镜观察细胞形态学改变和计数法测定生长曲线；用MTT比色法检测给药后鼻咽癌细胞的增殖抑制情况；采用流式细胞术和PI/Hoechst33258荧光双染法检测凋亡。结果 黄芩苷作用后,癌细胞生长延缓并萎缩，胞质粗糙，有大量颗粒状物堆积，而且药物浓度越大，形态学改变越明显；黄芩苷对CNE-2Z细胞的增殖和生长有显著的抑制作用并呈明显的时间-剂量依赖关系；流式细胞仪和荧光双染法结果提示黄芩苷可诱导CNE-2Z细胞凋亡。结论 黄芩苷能抑制CNE-2Z细胞的增殖效应，其机制可能与诱导细胞凋亡和引起细胞周期阻滞于G2/M期有关。

The inhibitory effect of baicalin on human nasopharyngeal carcinoma CNE-2Z cells

Objective To study the inhibitory effect and potential mechanism of baicalin on human nasopharyngeal carcinoma CNE-2Z cells.Methods The cellular morphology was observed by invert microscope.The growth curve was drawn by means of cell culturing and counting cell number.MTT assay was adopted to determine the proliferation of CNE-2Z cells.The cell apoptosis was detected by flow cytometery and Hoechst3358/PI staining.Results After baicalin treated,CNE-2Z cells grew slowly and atrophically.The severity of morphologic change was accompanied with baicalin dosage.The proliferation and growth of CNE-2Z cells were significantly suppressed by baicalin in time and does-dependent manner.The apoptosis of cells were determined by flow cytometery and Hoechst 3358/PI staining.Conclusion Baicalin can inhibit the proliferation of CNE-2Z cells.The possible mechanism may be related to inducing the cell apoptosis and arresting the cell cycle in G2/M phase.
