P物质抑制剂辣椒素和bFGF抗体对增生性瘢痕成纤维细胞增殖协同抑制作用的实验研究

摘    要:目的:探讨P物质抑制剂-辣椒素及bFGF抗体单独或联合使用对体外培养的人体增生性瘢痕成纤维细胞增殖的影响。方法:体外培养12例增生性瘢痕组织的成纤维细胞,分别加入不同浓度的辣椒素或/和bFGF抗体,培养24h后观察细胞形态学变化,并用MTT法检测细胞增殖抑制率。结果:所有不同浓度的辣椒素(2.5,5,20,40,60,80mg/L)均能使细胞皱缩、坏死,抑制细胞增殖,其抑制作用随剂量增大而增加。中高浓度bFGF抗体(40,80mg/L)也可促使细胞皱缩、坏死,抑制细胞增殖,其抑制作用也随剂量增大而增加。辣椒素与bFGF抗体联合应用对成纤维细胞抑制作用较单独使用效果更显著(P﹤0.05)。结论:辣椒素及bFGF抗体可抑制增生性瘢痕成纤维细胞增殖,其抑制作用随浓度增加而增加,两者联合使用有协调作用,本研究结果可能在增生性瘢痕的治疗提供新的思路和手段。

Abstract:Objective The present study was done to observe the effects of inhibitory substance P (capsaicin) and anti-basic fibroblast growth factor antibody on the fibroblasts derived from patients with hypertrophic scars, and explore their potential application in treatment of hypertrophic scar. Methods Hypertrophic scar-derived fibroblasts from 12 patients were harvested and cultured in vitro. After the cells were treated with different concentration of capsaicin and bFGF antibodies for 24 hours, the cellular morphology and proliferation were measured by microscope or MTT method, respectively. Results The cultured fibroblasts were shrinked by capsaicin (5, 20, 40, 60, 80mg/L) or anti-bFGF antibody (40, 80mg/L). Moreover, capsaicin at 5, 20, 40, 60, 80mg/L or anti-bFGF antibody at 40, 80mg/L significantly inhibit the proliferation of hypertrophic scar fibroblasts. Capsaicin at a dose 2.5mg/L or anti-bFGF antibody at a dose 20mg/L did not significantly inhibited cell proliferation (P>0.05), whereas co-administration of Capsaicin and anti-bFGF antibody at these doses significantly increase inhibitory effect on cell proliferation (P<0.05). Conclusion The proliferative activity of hypertrophic scar-derived fibroblasts could be inhibited by capsaicin or /and bFGF antibodies, in a dose - dependent manner. Coadministration of Capsaicin and anti-bFGF antibodies might be potential for the prevention and treatment of hypertrophic scars.
