海人酸诱导大鼠海马nestin的表达

摘    要:
为了探讨海人酸(kainicacid,KA)侧脑室注射后大鼠海马内nestin的表达变化,本文选用成年雄性SD大鼠,并随机分成实验组和对照组。实验组大鼠侧脑室注射1μl(0.5μg/μl)KA,分别于术后1、3、7、14、30、60、90d处死动物,并应用免疫荧光方法观察神经前体细胞标记物nestin、神经元特异烯醇化酶(NSE)和胶质纤维酸性蛋白(GFAP)的表达。实验组大鼠海马的nestin阳性细胞在KA注射后3d开始出现于齿状回、CA3和CA1区,到7d达到高峰,然后逐渐减少。注射侧的nestin阳性细胞数均较非注射侧多(P<0.01);对照组仅有散在阳性细胞。本结果提示,海人酸可诱导和增强大鼠海马nestin的表达,而这些表达nestin的细胞主要为星形胶质细胞。

Abstract:
To explore the change of the expression of nestin in the rat hippocampus after infusion of kainic acid (KA) into the lateral ventricle. Adult male SD rats were used and divided into control group and experimental group randomly. Rats in experimental group were infused with 1μl (0.5 μg/μl) KA into the lateral ventricle and sacrificed 1, 3, 7, 14, 30, 60, 90 days after infusion. The expression of nestin, neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) were observed by immunofluorescent staining. The nestin-positive cells in the experimental group appeared in dentate gyrus, CA3, CA1 on the third day and reached maximal expression level on the seventh day after infusion of KA and then decreased gradually. The number of the nestin-positive cells in the infusion side was more than that in opposite side (P<0.01) and there were seldom nestin-positive cells in the control group. The present study suggests that KA may induce and promote the expression of nestin in rat hippocampus and most of the nestin-positive cells were astrocytes.
