Abstract:Objective:To investigate the mobile phases influence on relative molecular weight(Mr) determination of Grifola Frondosa Polyaccharide(GFP) by High performance gel permeation chromatography(HPGPC). Methods: Determine Mr of GFP by HPGPC using different mobile phases, concentrations of NaC1 and pH value of GFP separately. Results:The Mr of GFP are 822410,177520,29112 and 25066 dalton determined by HPGPC using pure water,0. 02%NAN3 solufion,0.1mol·L-1 NaNO3 solution and 0. 1mol·L-1 Na2SO4 solution as mobile phase respectively. The Mr of GFP was sharply dropped under the eoneentration of 0 - 0. 025 mol/L NaCl and then tend to be stable from 0. 1 to 0. 2 mol/L NaC1. The Mr increase quickly from pH 3 to 6 ,and then maintain stable from pH 6 to 9 ,but slightly inereased at pH 10. Conclusion:Different mobile phases, concentrations of NaCl and pH value have great influence on Mr deterroination of GFP.
