摘 要: | 目的建立宣木瓜药材中绿原酸、芦丁、金丝桃苷、槲皮素的含量测定方法。方法采用HPLC法,Phenomenex luna C18色谱柱(250 mm×4.6 mm,5μm),流动相为0.6%磷酸溶液(A)和乙腈(B),梯度洗脱,检测波长为360 nm,流速设为1.0 ml.min-1。结果绿原酸、芦丁、金丝桃苷、槲皮素线性范围分别为0.035~0.56 g·L-1(r=0.997 8),0.003 2~0.051 2 g·L-1(r=0.999 3),0.004 25~0.068 g·L-1(r=0.999 7),0.003 5~0.056 g·L-1(r=0.999 7),平均加样回收率分别为102.9%(RSD=1.04%)、99.3%(RSD=2.28%)和99.2%(RSD=2.33%)、99.5%(RSD=2.52%)。结论该方法操作简单,结果准确,具有较好的重复性和稳定性,可用于宣木瓜药材的质量控制。

Abstract: | Objective To establish an HPLC method for the simultaneous determination of chlorogenic acid,rutin,hyperoside and quercetin in Fructus Chaenomelis.Methods Ahigh-performance liquid chromatography equipped a Phenomenex luna C18(250 mm×4.6 mm,5 μm) column with UV detection used.The mobile phase consisited of 0.6% aqueous phosphoric acid(A) and acetonitrile(B) with gradient elution.The detection wavelength was set at 360 nm and the flow rate was 1.0 ml·min-1.Results The linear ranges of chlorogenic acid,rutin,hyperoside and quercetin were 0.035~0.56 g·L-1(r=0.997 8),0.003 2~0.051 2 g·L-1(r=0.999 3),0.004 25~0.068 g·L-1(r=0.999 7),0.003 5~0.056 g·L-1(r=0.999 7),respectively;the average recoveries were 102.9%(RSD=1.04%),99.3%(RSD=2.28%),99.2%(RSD=2.33%),99.5%(RSD=2.52%).Conclusion The proposed method is simple,accurate,and repeatable.which can be used for quality control of Fructus Chaenomelis. |
