| 急性白血病IgH基因重排的研究 | |

| 摘 要： | 对１０５例儿童白血病细胞作了ＩｇＨ基因重排半巢式聚合酶链反应（ＰＣＲ）分析，结果显示重排主要见于Ｂ细胞系急性淋巴细胞白血病（阳性率７５．５％）及相关杂合性白血病（ＨＡＬ）中，说明ＩｇＨ基因重排ＰＣＲ分析结果可作为分型指标之一。１５例正常外周血淋巴细胞则无明显条带，说明可以区分克隆性和反应性增生。２２例同时与ＤＮＡ印迹比较，结果相关。５例ＰＣＲ产物序列分析表明序列同源性小，是克隆特异性的。３例动态分析示３例均在完全缓解期测得白血病克隆，说明ＩｇＨ基因重排ＰＣＲ分析可以检测白血病克隆的消长、演变，值得进一步研究。 |

| Abstract: | he DNAs from leukemic cells in 105 childrenwith acute leukemia were analyzed by using PCRmethod. The results indicated that IgH gene rearrange-ments were mainly in B precursor acute leukemia , sug-gesting that it can be used for leukemia subtyping. ThePBL DNAs from 15 normal subjects were also analyzedin the same way; however, no sharp band was noted ,which was different from those of leukemia. IgH generearrangements were further analyzed in 22 cases of a-cute leukemia by Southern blot. The resuhs obtainedby both methods showed a good correlation. Analysisof DNA sequences of PCR product was done in fivecases. The data showed that sequences of V-D-J rear-rangement was of clonal specificity. Dynamic analysisin totally three cases showed a clonal remission. It isconcluded that PCR analysis of IgH gene can be used todetect clonal changes in acute leukemia. |
