黄芩苷对糖尿病大鼠肾脏细胞凋亡及其Bcl-2和Bax表达的影响

摘    要:目的:研究黄芩苷对糖尿病大鼠肾脏细胞凋亡的影响,并从抗凋亡的角度探讨其作用机制。方法:采用小剂量高脂高糖喂养联合链脲佐菌素单次注射法造成2型糖尿病大鼠模型,黄芩苷[150mg(kg·d)]灌胃8周,8周后称体重、肾重,计算肾指数;收集大鼠血液和24h尿,测定血糖和尿微量蛋白;一侧肾脏采用TUNEL法检测凋亡及免疫组化检测Bcl-2和Bax蛋白的表达,另一侧肾脏液氮保存后采用荧光实时定量RT-PCR法测Bcl-2和BaxmRNA的表达。结果:与对照组比较,糖尿病组肾脏凋亡细胞数明显增多。Bax和Bcl-2表达均增强,Bax/Bcl-2增高(P<0.01);黄芩苷治疗组较糖尿病组凋亡细胞数减少,Bax表达减弱,Bcl-2表达增强,Bax/Bcl-2降低(P<0.01)。结论:黄芩苷可以抑制糖尿病大鼠肾脏细胞凋亡,其作用机制可能与影响凋亡基因Bax和Bc1-2表达有关。

Abstract:Objective To investigate the effects of baicalin on the renal cell apoptosis in the diabetic rats.Methods Diabetic rat models were induced by high-lipids and high-sucrose feeding for a month and single intraperitoneal injection of streptozotocin(STZ),then were administrated with 150 mg/(kg·d)of baicalin for 8 weeks.At week 8,the body weight and renal weight were measured and then the renal index was calculated.At the end of the experiment,the blood and 24h urine samples were collected for determination of blood glucose and urine protein.The apoptosis was detected by terminal deoxynucleotidyl transferase mediated nick end labeling(TUNEL),and the expressions of Bcl-2 and Bax proteins were detected by immunohistochemical methods in one side of kidneys.The expressions of Bcl-2 and Bax mRNAs were detected by fluorescence real-time quantitative RT-PCR in another side.Results As compared with those in control group,the number of apoptotic cells,expressions of Bax and Bcl-2,and Bax/Bcl-2 ratio were significantly increased in diabetic group(P < 0.01).By baicalin treatment,the number of apoptotic cells and the expressions of Bax and Bax/Bcl-2 ratio were decreased(P < 0.01),while Bcl-2 was increased(P < 0.01)as compared with those in diabetic group.Conclusion Baicalin may inhibit the apoptosis of renal cells in diabetic rats by regulating the expressions of Bcl-2 and Bax.
