摘    要:
目的:探讨FAF1表达在人胃癌发生发展中的作用。方法:应用实时荧光定量PCR(FQ-RT-PCR)和免疫组化方法,对40例胃癌患者癌组织、癌旁组织及相对正常组织中FAF1mRNA和蛋白的表达进行分析。结果:FAF1mRNA在胃癌组织中的表达水平(0.27±0.12),明显低于在癌旁组织(0.40±0.06)及正常组织(0.48±0.08)中的表达水平(P<0.01),而在癌旁组织中的表达水平明显低于正常组织中的表达水平(P<0.01)。胃癌组织的FAF1蛋白的阳性表达率(37.50%)明显低于癌旁组织(62.50%,P<0.05)和正常组织(72.50%,P<0.01)。结论:FAF1表达的减少在胃癌的发生发展过程中起着重要作用。

Abstract:
Objective To investigate the relationship between Fas-associated factor 1 (FAF1) expression and gastric mucosa carcinogenesis.Methods The mRNA and protein levels of FAF1 in 40 cases of gastric cancer,paracancerous and normal tissues were analyzed by fluorescent quantitative real-time polymerase chain reaction (FQ-RT-PCR) and Immunohistochemistry respectively.Results The mRNA levels of FAF1 in the cancerous tissues (0.27 ± 0.12) were significantly lower than those in the paracancerous tissues (0.40 ± 0.06) and normal tissues (0.48 ± 0.08)(P < 0.01).And the protein level of FAF1 in the gastric cancerous tissues (37.50%) were also significantly lower than those in the paracancerous tissues (62.50%,P < 0.05) and normal tissues (72.50%,P <0.05).Conclusion Downregulated expression of FAF1 may play an important role in the occurrence and development of gastric cancer.
