摘    要:
建立大鼠血浆中吴茱萸次碱的反相高效液相色谱法。以乙腈 水 (5 8∶4 2 )为流动相 ,地西泮为内标 ,在波长 3 4 5nm处检测。吴茱萸次碱浓度在 0 0 2~ 1 6μg/mL呈线性 ,相关系数为 0 999,高中低 3种浓度平均萃取回收率均大于 80 0 %

Abstract:
An accurate, convenient and sensitive HPLC method with UV detection was established to determine the rutaecarpine in the rat plasma. The rutaecarpine was determined by RP HPLC.The pretreatment of the plasma sample was based on single liquid liquid extraction. Chromatographic conditions included mobile phase acetonitrile water(58∶42), internal standard diazepam, detector UV, 345 nm . The working curve was linear in the concentration range of 0 02~ 1 6 μg/mL . The average recoveries of high,mid and low concentrations of rutaecarpine were higher than 80 0%. This method was used for determination of the concentration of rutaecarpine in the rat plasma. It is satisfactory for the pharmocokinetics study of rutaecarpine.
