摘    要:目的高速逆流色谱法分离纯化小花黄堇中延胡索乙素和原阿片碱。方法采用高速逆流色谱法进行分离,以正已烷.乙酸乙酯.甲醇.水(8:8:12:8,V/V)为溶剂系统.上相为固定相,下相为流动相,转速:800r·min一,流速:2mL·min-1,检测波长:280nm。结果从小花黄蔓总生物碱提取物中分离得到延胡索乙素和原阿片碱。纯度分别可达99.2%和99.6%。结论利用高速逆流色谱法可高效分离和纯化小花黄堇中延胡索乙素和原阿片碱。

Abstract:OBJECTIVE To separate dl-tedrahypalmatine and protopine from Corydalis racemosa(Thunb.) Pers.by high-speed counter-current chromatography(HSCCC).METHODS Using high-speed counter-current chromatography,solvent systems was composed of n-hexane-ethylacetate-methanol-water(8∶8∶12∶8,v/v),and its upper phase was used as the stationary phase and its lower phase as the mobile phase,separation was performed at the rotating speed of 800r·min-1,flow rate of 2.0mL·min-1 and detection wavelength of 280nm.RESULTS Dl-tedrahypalmatine and protopine from Corydalis racemosa(Thunb.) Pers.were isolated with the purity of 99.2% and 99.6%.CONCLUSION Dl-tedrahypalmatine and protopine from Corydalis racemosa(Thunb.)Pers.could be separated and purified by HSCCC with high-efficiency.
