一侧颈总动脉结扎对小鼠脑含水量和血管内皮生长因子表达的影响

摘    要:目的:观察一侧颈总动脉结扎(uCCAO)后小鼠脑含水量和血管内皮生长因子(VEGF)的表达情况。方法:116只雄性昆明小鼠,随机分成假手术组和uCCAO组各58只。uCCAO组结扎左侧颈总动脉,假手术组不结扎。干湿重法测量小鼠术后第1、3、5、14天脑组织含水量。术后第14天脑组织切片行HE染色、VEGF免疫组织化学染色和Western Blot。结果:术后笫3天uCCAO组小鼠脑含水量高于假手术组(P<0.05)。HE染色显示uCCAO组小鼠脑组织形念结构基本正常。免疫组织化学染色示uCCAO组和假手术组小鼠左侧脑组织均可见VEGF阳性细胞。Westem Blot示uCCAO组小鼠左侧皮质VEGF表达较假手术组显著增加(P<0.05)。结论:昆明小鼠uCCAO可以造成脑缺血改变,并首次发现小鼠uCCAO脑缺血模型急性期脑组织VEGF表达上调。

Effects of Unilateral Common Carotid Occlusion on Brain Water Content and VEGF Expression in Mice

Abstract:Objective:To observe the brain water content and the expression of vascular endothelial growth factor(VEGF) in the brain of mice after unilateral common carotid occlusion(uCCAO). Methods:One hundred and sixteen Kunming male mice were randomly assigned to the sham group and uCCAO group equally.The left common carotid arteries were separated and ligated in the uCCAO group,whereas the arteries were separated but not ligated in the sham group.The brain water content was measured on the 1st,3rd,5th,and 14th day after operation with the dry-wet method.Mice were harvested on the 14th day for the HE staining,vascular endothelia growth factor(VEGF) immunohistochemical labeling and western blot.Results:Brain water content was increased significantly in the uCCAO group on day 3 when compared with the sham group(P<0.05).HE staining was normal in the uCCAO group.The immunohistochemical labeling showed the presence of VEGF-positive cells in the left hemisphere in the both groups on day 14.Western blotting demonstrated that the expression of VEGF in the cortical areas was significantly increased in the uCCAO group when compared with the sham group(P<0.05).Conclusion:uCCAO operation results in brain ischemic changes in Kuming mice,and the uCCAO brain ischemia model up-regulates VEGF expression in the brain of mice at the acute phase.
