益骨胶囊对去卵巢大鼠骨密度及血浆和骨组织CGRP和SP含量的影响

摘    要:目的观察补肾活血中药复方益骨胶囊对去卵巢大鼠骨密度及血浆和骨组织中降钙素基因相关肽(CGRP)、P物质(SP)含量的影响,探讨其作用机制。方法 6月龄SD雌性大鼠40只,随机分为4组:假手术组、模型组、益骨胶囊低剂量组、益骨胶囊高剂量组。模型组、益骨胶囊低剂量组、益骨胶囊高剂量组均行双侧去卵巢手术,术后1周益骨胶囊低、高剂量组用灌胃法给服益骨胶囊(1.16 g/100 g和10.44 g/100 g),模型组灌服生理盐水,12周后观察各组大鼠骨密度的改变,并用ELISA法检测各组大鼠血浆和骨组织CGRP和SP的含量。结果与假手术组比较,模型组骨密度显著降低(P<0.01),血清和骨组织中CGRP和SP也明显降低。与模型组比较,益骨胶囊低剂量和高剂量组骨密度明显增高(P<0.01),血浆和骨组织CGRP和SP明显增高(P<0.01),但仍低于假手术组(P<0.01)。结论益骨胶囊能够调节血浆和骨组织CGRP和SP的含量,这可能是其抗骨质疏松的作用机理之一。

Abstract:Objective To investigate the effects of Benefiting-bone capsule(BBC) on bone mineral density and the levels of CGRP and SP in plasma and bone tissue in ovariectomized rats,and to further explain the mechanism of BBC on preventing and treating osteoporosis.Methods 40 SD rats were randomly divided into four groups:sham-operated group,model group,BBC low-dose group and high-dose group.After ovariectomy 1 w,BBC groups were given by intragastric administration with different doses of BBC and model group was given with normal physiological saline.After 12 w,the change of bone mineral density were observed,the levels of CGRP and SP in plasma and bone tissue were measured by ELISA.Results Compared with sham-operated group,BMD of model group was decreased significantly(P<0.01),the levels of CGRP and SP in serum and bone tissue were also decreased significantly(P<0.01).The bone mineral density in BBC low dose group and BBC high dose group was significantly higher than that of model group(P<0.01),the levels of CGRP and SP in plasma and bone tissue was significantly higher than those of model group(P<0.01),but still lower than that of the sham-operated group(P<0.01).Conclusion BBC can regulate the levels of CGRP and SP,which may be one of its mechanisms of anti-osteoporosis.
